fullyrespiratorygalactosemetabolismreliesexclusivelyonthePEP-glyoxylatecycle,incontrasttothecanonicalrespiro-fermentativegrowthonglucose.Alarge-scale13C-fluxanalysisof91transcriptionalregulatormutantsonbothsubstratesdemonstratedtheimportanceoftranscriptionalregulation,with2/3oftheregulatorsdirectlyorindirectlyaffectingabsolutefluxes.Ourdatademonstrated,however,thatthefluxpartitioningbetweendifferentpathwaysremainedlargelystable,withonlytheacetyl-CoAbranchpointbeingsubjecttogeneticcontrol.Thus,weprovidestrongevidencethattranscriptionalregulationinE.coliisprimarilyinvolvedincontrollingthemagnitudeoffluxthroughmetabolicpathwaysbutrarelythedecisionoffluxdistributionbetweenpathways.Inlinewiththeemergingviewthatthemetabolomeisanactiveplayerinregulatingfluxthroughcentralmetabolism,wesystematicallyinvestigatedtowhichextentthemetabolomemightplayaroleinfluxregulationinChapter3.Byinferringdistributionsofintracellularfluxesusing13C-wholeisotopologuebalancingandbyquantifyingintracellularmetabolitesupon8carbonsourceperturbations,weconfirmedknownmetabolitereportersandhypothesizedpotentialreportingby3others.Rathersurprisingly,wefoundabout2/3ofthe41quantifiedintracellularmetabolitestocorrelatewithfluxmagnitudeand/orfluxusage,renderingthematleastpotentialcandidatesforputativefluxreporting.WhetherornotthisreportingpotentiallyisactuallyusedbyE.coliremainstobetested.However,theresultsdemonstratedthepotentialoffutureresearchinsearchingforadditionalallostericmetabolite-proteininteractions.Abstract9Averydifferentquestionrelatedtothedistributionoffluxesishowcellsactuallycontroltheirnetwork-widebalancingofcatabolicNADPH-producingandanabolicNADPH-consumingfluxes?
InChapter4wedemonstratedthattranscriptionalregulationofthetwotranshydrogenasesdoesnotresponddirectlytotheintracellularredoxcofactorrequirements.Instead,byidentifyingthetranscriptionfactorsthatregulatetranscriptionofbothisoforms,weshowedthatthepntABandudhApromotersrespondtosignalsthatinfluencetheNADPHbalance(morespecificallythepresenceofaminoacidintheenvironment,oxidizedquinonesandconditionsofhighgrowthrate).Additionally,ourresultsdemonstratedthatatleastonefurtherregulationmechanismmustexist.ThereforethetranshydrogenaseisoformsinE.colimightachievethenetwork-widebalancingofcatabolicNADPH-producingandanabolicNADPH-consumingfluxesviaotherregulatorymechanisms,i.e.allosteric/kineticcontrol.Finally,toachieveamorecomprehensiveunderstandingoftheoverallimportanceoftranscriptionalregulationofcentralmetabolicflux,weconductedaregulationanalysisontranscriptandfluxdataforeightcarbon-sourceperturbationsinChapter5.Wedemonstratedthatonaverage16%ofthefluxchangescanbeexplainedthroughthecorrespondingchangesinmRNAlevels,whichwassurprisinglyconsistentwithaprevious,purelycomputationalanalysis.Withinmetabolicpathwaysthecontributionoftranscriptionalregulationtofluxregulationvariedsubstantially,illustratingacomplexregulationpattern.Alsoacrossconditionsthecontributionoftranscriptionalregulationforregulationoffluxthroughenzymesvariedsubstantially.OverallourresultsinChapter5emphasizedamuchstrongerrolefortranscriptionalregulationoffluxinE.coliwhencomparedtothereportedvaluesinSaccharomycescerevisiae.However,theresultsalsodemonstratedsubstantialcontributionoffluxregulationtooccurviaotherregulatorymechanisms.Abstract10ZUSAMMENFASSUNGStoffwechselversorgtZellenmitBausteinenundEnergiefrallezellulrenProzesseundermglichtdadurchLebenundProliferationinsmtlichenOrganismen.DasVerstndnisdesStoffwechselsundseinerRegulationistvonzentralerBedeutungnichtnurfrDiagnoseundTherapievonStoffwechselerkrankungen,sondernauchfrdiemikrobielleProduktionvonChemikalien.DieIdentifikationderProzesse,diemetabolischeFlsseimZentralstoffwechselaktivregulieren,isteinwichtigesForschungsgebietdermikrobiellenSystembiologie.InKapitel2dieserArbeituntersuchtenwirdieerstaunlichgrossenUnterschiedeimStoffwechseldersehrhnlichenSubstrateGalaktoseundGlukoseinEscherichiacoli:
